GenomONE™-Neo EX 仙臺(tái)病毒包膜轉(zhuǎn)染試劑
可用于DNA、siRNA、ODN及蛋白轉(zhuǎn)染!
◆原理:
待轉(zhuǎn)移的分子(如DNA,蛋白,Antisense Oligonucleotide, siRNA等)與HVJ-Envelope (HVJ-E) 混合后,形成了HVJ-E載體。利用病毒衣殼蛋白的侵染能力,在融合蛋白(F)的作用下,直接與細(xì)胞融合,將分子轉(zhuǎn)入目標(biāo)細(xì)胞或組織。

2. 安全性
與陽(yáng)離子脂質(zhì)體不同,GenomONE-Neo是通過(guò)膜融合作用將分子導(dǎo)入細(xì)胞,而脂質(zhì)體轉(zhuǎn)染的內(nèi)吞作用可能造成轉(zhuǎn)染分子經(jīng)溶酶體作用而降解。GenomONE-Neo 內(nèi)含完全滅活的HVJ病毒顆粒,其具有侵染力而無(wú)潛在增殖能力,使用非常安全,無(wú)需特殊的防護(hù)措施

【大鼠頜下腺逆行注射siRNA】
Ca2+-dependent Cl channel protein (rCLCA)表達(dá)的特異性抑制。
免疫染色
non-injection side rCLCA siRNA-injection side
rCLCA siRNA(2 nmol)和GenomONE-Neo(2AU)混合后,以逆行注射的方式注入大鼠頜下腺。四十八小時(shí)后,用免疫染色檢驗(yàn)siRNA注射對(duì)氯離子通道蛋白表達(dá)的抑制作用。

■ in vitro
【難轉(zhuǎn)染細(xì)胞——U937細(xì)胞RNAi】
總所周知,用脂質(zhì)體和電轉(zhuǎn)的方法將siRNA轉(zhuǎn)到U937人單核細(xì)胞白血病細(xì)胞中非常困難,但我們通過(guò)實(shí)驗(yàn)證明,siRNA與GenomONE-Neo結(jié)合后轉(zhuǎn)染該細(xì)胞,其knock-down效率可達(dá)85%以上。
♦ U937細(xì)胞經(jīng)siRNA轉(zhuǎn)染72小時(shí)后收集,用RT-PCR和WB分別檢測(cè)mRNA和蛋白量的變化情況。
