1. 采集后長期保存的血液作為模板,能進行PCR 嗎?
實驗證明,采集后凍存了長達五年時間的血或紙血(干燥血)作為模板,依然能進行PCR。必須長期穩(wěn)定保存血液時,可使用Whatman 公司的FTA Card(核酸保存用卡)進行保存。關于血液(全血)對PCR 的抑制作用,直接采集的血液(新鮮血)對PCR 抑制作用更強。
另外,長久以來的經(jīng)驗證明,采用干燥血作為PCR 模板時,對比用一般的實驗紙保存,使用FTACard 保存的血液進行PCR 的結果更穩(wěn)定,因此推薦使用FTA Card 保存的血作為PCR 模板。
2. 從糞便中抽提的DNA 模板,可用PCR 檢測出細菌嗎?
糞便抽提的DNA 對PCR 有抑制作用,因此用稀釋后的DNA 反應。從糞便樣品中簡便進行PCR 的應用例子,在“使用實時PCR 法高靈敏度檢測Vero Toxin”中有介紹。將10%的糞便懸濁液熱處理(95℃,5 分鐘)后,將離心后的上清液作為PCR 模板。實驗證明,使用這個方法,大部分的糞便樣品都能穩(wěn)定進行PCR。
3. Ampdirect 可應用于RT-PCR 嗎?糞便里存在的RNA 病毒也可以檢測出來嗎?
從糞便樣品中簡便檢測RNA 病毒的應用例子,可見“糞便中的RNA 病毒檢測”。將糞便懸濁液離心后的上清液,涂布在Whatman 公司的FTA Card(核酸保存用卡)上,干燥后,在卡上打(φ1.25mm)就可作為模板使用。
Ampdirect Plus 可做為逆轉錄反應的反應液,逆轉錄酶選用的是Invitrogen 公司的M-MLVReverse Transcriptase(Cat No. 28025-013)。
4. 樣品的溶解處理最后步驟要在95℃,進行5 分鐘的熱處理,這個熱處理步驟是必須的嗎?
因為要使溶解液中的Proteinase K 失活,所以必須熱處理。如果不進行熱處理,Proteinase K 仍保持活性的話,在往PCR 反應液里加入溶解液時,Proteinase K 會將PCR 酶(Taq DNA Polymerase)降解掉,從而不能進行穩(wěn)定的PCR。實際上,這是從大部分得不到目的PCR 產(chǎn)物的例子上取得的經(jīng)驗。所以,必須在最后對溶解液進行熱處理(95℃,5 分鐘)。
5. 從植物的葉片抽提的DNA 作為PCR 模板進行反應,但得不到目的PCR 產(chǎn)物。考慮到是植物里所含的糖類抑制了PCR,使用Ampdirect 可以改善這個情況嗎?
至今為止,已有非常多客戶反映,從實驗植物、谷物、蔬菜、果蔬的葉片取樣,用Ampdirect進行PCR,都取得了良好的結果。不同植物對PCR 的抑制作用也不盡相同,可參照“從植物取樣的簡便PCR 實例”。在PCR 前用溶解液進行前處理。如擔心因為PCR 被抑制而得不到目的PCR 產(chǎn)物,用溶解處理液稀釋5~10 倍,會對此情況有很大改善。
6. 小鼠尾部用溶解液溶解了大約1 個小時,還沒完全溶解。可作為PCR 的模板使用嗎?
將小鼠尾巴1~5mm 浸入100μL 溶解液內(nèi),55℃下溶解約1 小時后,如小鼠尾巴還未完全溶解,對Proteinase K 進行失活處理(95℃,5 分鐘)后,從溶解液中提取出來的DNA 量(拷貝
數(shù))應該足以作為PCR 模板,大部分情況下進行PCR 沒有問題。
與此對照,如果是將大條的小鼠尾巴(5mm 以上)過夜處理以至完全溶解,溶解液會變成高粘度狀態(tài)。這種情況下,過剩的DNA 反而會抑制PCR 反應,請用蒸餾水或TE 將溶解液稀釋10 倍左右。請注意盡量不要讓小鼠尾巴過度溶解。
另外,溶解處理后的溶解液不需離心,只需取上清液就可作為PCR 模板使用。從以往經(jīng)驗得知,經(jīng)溶解處理過的溶解液在冷藏保存條件下保存2~3 年內(nèi)皆可作為PCR 模板使用。
7. 不對唾液進行前處理就直接作為PCR 模板是不可行的吧?“取樣自口腔粘膜的PCR”這個應用,是對口腔黏膜細胞用溶解液進行溶解處理,如果是用唾液的話,也必須用溶解液進行溶解處理嗎?
目前,也有不少客戶為了檢測唾液中的微生物,不進行前處理,直接將唾液當成PCR 模板使用。當作為模板進入PCR 反應液的唾液中(數(shù)μL)有大量微生物,可獲得良好的PCR 產(chǎn)物。與之相反,若微生物數(shù)量較少,根據(jù)唾液中的蛋白等物質對PCR 抑制的不同程度,有可能得不到穩(wěn)定的PCR 產(chǎn)物。因此,推薦對溶液用溶解液進行溶解處理,再作為PCR 模板。對唾液進行了溶解處理的話,可得到以下兩種結果,①“抽提微生物的DNA”和②“降低唾液對PCR 的抑制作用(分解唾液中的蛋白)”,也可得到穩(wěn)定的PCR 結果。
8. 如何用PCR 檢測血漿、血清樣品中的微生物?
血漿、血清這些全血,對PCR 有較強抑制作用,所以不能將全血直接用作PCR 模板。因此,要用于“檢測血清中的病毒”時,將血漿、血清用溶解液進行溶解處理,分解掉血漿、血清中的蛋白,降低對PCR 的抑制作用。
請注意,本方法適用于檢測DNA 病毒、細菌,不可用于檢測RNA 病毒。檢測RNA 病毒時,在溶解處理步驟,因為血漿、血清中存在RNA 分解酶,從病毒外殼裸露出來的RNA 就會被分解。
9、使用FTA Card 和實驗紙作為模板進行多樣品PCR 時,因為使用打孔器等工具可能會引起污染。有沒有相應的對策呢?
采集血液樣品時,首次打孔取樣后,在下一次取樣前,用打孔器在FTA Card 或實驗紙上沒有血樣的空白處空打3 個孔,這樣可保證打孔器上不會殘留前一樣品的模板(血樣是白血球DNA)。因為從客戶處得知,“每次用打孔器取樣時,用金伯利(Kimwipe)浸漬酒精擦拭打孔器的打孔部分,這個方法反而引起污染”,所以推薦空打孔的方法。但是,檢測拷貝數(shù)多的DNA,如線粒體DNA 和質粒DNA 等時,本方法不能防止污染。
